I bough this subscription , these videos are from appdev training and they are of high quality.You will get all the new videos for free during your time of subscription. Current training , I find really useful are :

For the full list , you can find it here. They provide trial as well, so you can watch some sample training before you buy.The only downside I find is that , the training videos are streamed from the internet and no download.It is kind of problem after 'Big time' is gone. But given the price is so low,it is still good bargain.

It really depends on the business case.You can do a lot of cool stuff without using UpdatePanel in MS Ajax.JQuery is just library which Microsoft adopted recently (actually a year ago) in its Microsoft ajax offering.
